The visitors pressed home their advantage building up a first innings lead of 382 after being bowled out for 592 thanks to hundreds from Rikki Clarke and Rory Burns.
Mullaney remains at the crease with the only Notts victim of the day being Jake Libby, who was pinned in front of his stumps by Morne Morkel, with the home side ending the day on 57-1.
